The clearest current human evidence is still in knee osteoarthritis, where glucosamine has mixed signals for pain and stiffness and a separate, cautious signal for slower worsening on X-rays in some long-term studies. Results vary a lot by formulation: some glucosamine sulfate studies look better than glucosamine hydrochloride or glucosamine-plus-chondroitin combinations, and none of this proves cartilage regrowth or a cure.
